1014 Novel mechanisms underlying Asian atopic dermatitis (AAD) highlight key functional networks in children identified by integrating untargeted skin proteomics and secretome

We aimed to decipher the key mechanisms involved in Asian Atopic Dermatitis using untargeted proteomics analysis of non-lesional skin and plasma Singaporean children. To identify active disease markers we used 3 groups children at year 10 age: those with (Active AD, N=20), history AD but not current (Past N=22) controls no allergies (Controls, N=29). Skin tape samples were processed by functional lab (FPL) Quad-orbitrap mass spectrometry which resulted 2468 protein IDs UniProt database. A Kruskal-Wallis test was carried out for significant differences (p<0.05) levels a minimum median difference 0.5 effect size. yielded total 246 proteins 40, 184, 164 Control vs Past (C/P), Active (C/A), PastAD (P/A) respectively, highlighted that major differentials from group. Key pathways includes S100 (S100A2, S100A7 S100A6) upregulated increased Keratin 16 (K1C16) 14 (K1C14), both undifferentiated keratinocytes. further observed upregulation Desmoglein-1 (DSG1) Desmoglein-3 (DSG 3). Taken together, having high keratinocytes causes an imbalance potentially resulting compromised barrier AD. Furthermore, downregulation loricrin (LORI), terminally differentiating structural filaggrin FLG, candidate gene integrity inflammatory signature plasma. Our study highlights novel known candidates immune dysregulation can be exploited biomarkers therapeutic intervention.
